La Salle College Antipolo's groundbreaking ceremonies, signaling the start of construction works, were held on 16 March 1985. The school's initial year suffered some setbacks brought about by the enormous cost of building a school atop a rocky mountain and by the impassable national road leading to the subdivision. In spite of these difficulties, classes in Kindergarten up to Grade Four went on temporarily at La Salle Green Hills in June until December. On 13 January 1986, classes were transferred to the original site composed of two buildings. The third building was completed in the summer of 1987, which was followed in 1990 by the High School building.
The College Department followed suit, offering Accountancy, Commerce, Psychology and Hospitality Management programs. A community of Brothers was also opened to fully supervise the entire operations of the college.
